Retrograde degeneration models

JSW has established a cell culture model specifically addressing retrograde degeneration of already formed neuronal processes. Adult dorsal root ganglia from rats are cultured as explants in vitro prior to lesion with:

• Vincristine sulphate
• Paclitaxel

These lesions evoke a die-back of extended neurites similar to neuropathies. Cultures are analysed for neurite length and area of neuritic halos by immunolabelling for neurite specific markers and semi-automatic quantification (see Analyses of culture models).
